研究内容要約 Research Summary |
多摩川の水質保全を流域住民の健康長寿の達成という視点から、①独自に開発した新しい水質検査法を用い、流域水道水も検査対象とし、②市民講座などの開催により水に関わる啓蒙活動と健康調査を行った。その結果、上述①の目的に適う培養ヒト細胞を開発できたことである。また、この細胞を用いての細胞増殖阻害度測定により、多摩川の水の阻害度は上流から下流へ下がるほど上昇し、その上昇と連動するかのように、流域水道水の水質悪化が高頻度で見られ、下流へ行くほど検出されやすいのではないかという疑問が生じたことである。一方、②については、9回の市民講座を開催した。その講座でのアンケート調査により、水と健康に関わる真摯かつ多様な要求のあることが判明した。そこで、市民や学生と共に開設する多摩川と流域住民の健康を守る教育アカデミーの設立を八王子市内で目指すこととし、その基盤を構築した。この水質悪化の連動性の可能性をさらに追究したが、水道法に基づく通常の水質検査法で水質悪化は確認されず、私共の独自の水質検査法の必要性が判明した。 The purpose of this study is to evaluate the biological effects of potable water from the Tama River system on human cells using bioassays and to construct the educational system which supplies information about the role of water on human health to citizens around the Tama River area For the former purpose, we established human E-RSa cells which contain c-DNA for estrogen-receptor gene and therefore produce the estrogen receptor protein from RSa cells . The parent RSa cells are highly sensitive to the genotoxicity of environmental chemicals , and the variant E-RSa cells are suitable for detecting estrogen-like activity of environmental chemicals Thus, the two cell lines were used for the bioassay. Water samples from the upper and lower reaches of the Tama River were collected . Tap water samples from Tama River system were also collected at different points close to the above Tama River area. Organic chemical compounds in the water samples were concentrated by adsorption to OASIS HLB resin and used for the bioassay. The concentrated samples from the lower reaches of the Tama River showed greater growth-inhibiting effects on the cells than those from the upper ones. The concentrated samples of tap water also showed more significant growth - inhibiting effects at lower parts than those at upper ones. For the latter aim of educational system construction, we gave lectures on life-water science at nine different halls, including collection of blood samples from more than 50 participants On analysis of the ability of human serum to regulate cell proliferation, cyto-toxic mutation-inducing activity was not observed in serum from any of those volunteers, who drink water from the Tama River system daily. However , all of the participants who corresponded to the questionnaires desired the sustainable citizen education . |
