堆積物から核酸を効率よく抽出し、精製する方法を検討し、確立した。この方法を用いて 、rRNA多摩川下流堆積物からを抽出し、全真正細菌、各種の硫酸還元菌に特異的なオリ ゴヌクレオチドプローブとハイブリダイゼーションを行ない、各種の基質を添加した堆積物に おける硫酸還元菌の動態を調べた。 乳酸とプロピオン酸添加では多摩川汽水域、海水域ともDesulfobulbusが優占し、酢酸で は海域でのみDesulfobacterが優占した。 また、数地点の堆積物で調査した結果、各種の硫酸還元菌が広く分布する既知の硫酸 還元菌のRNAの比率は小さいことが分かった。 We developed a method to effectively extract and purify nucleic acid from sediment. This method was used to analyze the population dynamics of sulfate-reducing bacteria in sediment to which several substrates were added. More specifically, we extracted rRNA from sediment in the lower Tamagawa river; the extracted rRNA was hybridized with oligonucleotide probes that are individually specific to all bacteria and various sulfate-reducing bacteria. After the addition of lactic acid and propionic acid, Desulfobulbus became dominant in both the estuary basin and the saltwater basin of the Tamagawa River . After the addition of acetic acid, Desulfobacter became dominant. The examination of sediment taken from several sites indicated that, in areas where diversified sulfate-reducing bacteria are widely distributed, there is only a small percentage of RNA derived from known sulfate-reducing bacteria. |
